Title
A genome-wide association study of non-Hodgkin lymphoma
Summary
Genetic factors have long been recognized as influencing the risk of non-Hodgkin lymphoma (NHL). Recent genome-wide association studies (GWAS) of follicular lymphoma (FL) and chronic lymphocytic leukemia (CLL) have identified susceptibility loci for these common NHL subtypes. It is expected that there are additional common risk alleles which could influence susceptibility to these and other NHL subtypes. To identify such variants, we propose to conduct a large-scale GWAS with the aim of identifying genetic markers associated with risk for the five most common NHL subtypes [FL, small lymphocytic lymphoma (SLL)/CLL, diffuse large B-cell lymphoma (DLBCL), marginal zone lymphoma (MZL), and mantle cell lymphoma(MCL)] in non-Hispanic Caucasian participants from case control and cohort studies (including PLCO) participating in the Interlymph Consortium and the NCI Cohort Consortium, respectively. Secondary aims of this GWAS include performing pathway analyses, investigations of gene-gene and gene-environment interaction, and analyses to identify markers associated with survival. Specifically with regard to PLCO, we seek permission to extract and genotype DNA from all incident NHL cases accrued in NHC PLCO participants (approximate N = 843). Non-Hispanic Caucasian PLCO subjects previously scanned as controls in previous GWAS projects with no history of NHL will be included in the statistical analysis as controls. We plan to scan at least 10,000 subjects using the Illumina® Human660W chip. Cases and controls that are not scanned will be genotyped by TaqMan for the most noteworthy findings that emerge from the genomewide scans to increase statistical power. These new data will be combined as appropriate with results from the initial scans of NHL currently in progress or completed by InterLymph investigators (involving 1,800 cases and 2,000 controls) and others. PLCO will be an extremely valuable contributor towards this project, given its large number of accrued NHL cases with DNA and thousands of previously scanned controls.
Aims

1) To conduct a genome-wide scan of NHL focusing on specific subtypes (FL, SLL/CLL, DLBCL, MZL,
MCL) using a subgroup of cases and controls from InterLymph case-control studies and cohort studies on the Illumina® Human660W, with follow-up genotyping of top findings in remaining cases and controls as needed to maximize the statistical power.
2) To conduct follow-up pathway, gene-gene, gene-environment and survival analyses through the InterLymph infrastructure (e.g., NHL GWAS Coordinating Committee, InterLymph Working Groups).
