Title
Pan-cancer genome-wide association study of tissue disruption phenotypes on standard histologic images from PLCO
Summary
Disruption of organ-specific tissue ecosystems is a hallmark of malignant transformation, tumor progression, and clinical outcomes, particularly for solid cancers. Germline genetic variants in susceptibility genes have been shown to be associated with specific pathological characteristics of tumors from several sites. However, it is not known whether germline genetic susceptibility variants confer vulnerability to, and/or impact the pattern and degree of, tissue ecosystem disruption in tumors. Here, we aim to characterize the relationships between germline genetic variation and organ-specific tissue ecosystem disruption phenotypes across several cancer sites in the Prostate, Lung, Colorectal, and Ovarian cancer (PLCO) screening trial.
Aims
i. Characterize tissue ecosystem disruption phenotypes in tumor tissues using computational pathology of standard histologic images
ii. Conduct a genome wide association study (GWAS) to test associations between germline genetic variants and distinct tissue disruption phenotypes in several cancers.
