Summary
Smoking and Lung Cancer in PLCO and NLST
Project Summary: The Lung U!9 (NIH U19 CA148127-01) and Genetic Epidemiology Branch investigators propose to look at NLST data to examine the joint effects of smoking and genetic exposures on lung cancer in a range of populations. We intend to compare allele frequencies and magnitude of disease risk for a variety of variants and smoking behaviors. Data will be examined in NLST, as well as other population-based cohorts such as ACS, PLCO, ATBC, Caret, and the Nurses Health Study.
We will consider smoking but also take into account key covariates such as age, family history and COPD. We will also conduct specific analyses in controls to look at more detailed smoking behaviors, and to construct and validate risk models that incorporate the effects of smoking and other environmental factors.
Aims

Specific Aims:
To compare lung cancer risk factors across NLST with other population-based cohorts including PLCO, Nurse Health Study, CARET, ATBC amnd others.
To compare heterogeneity of effects for key lung cancer risk variants by subject and disease characteristics
To examine lung cancer risk models incorporating genetic and environmental data.
