Title
Study of Aberrant Crypt Foci (ACF)

Summary
This study will enroll about 600 PLCO subjects from 4 centers to undergo two annual exams with high resolution endoscopy. The examiners will search for aberrant crypt foci (ACF) in the rectum and take biopsy specimens. The lab at UCLA will perform histological and other analyses on the biopsy specimens.

Aims

1. Assess aberrant crypt foci (ACF) prevalence and 12 month interval natural history (including ACF incidence) within a defined area of colorectal mucosa. 2. Evaluate the concordance between endoscopic and histopathologic characterizations of ACF. 3. Correlate changes in cellular/molecular (using multiplex tissue, genomic, and proteomic array technologies) biomarkers from normal-appearing mucosa and ACF with more advanced colorectal neoplasia (i.e., adenomas or cancer), if any are detected in the study population. 4. Evaluate the association between ACF status and several potential etiologic risk factors (i.e., age, gender, dietary factors, tobacco use, alcohol use, aspirin/NSAID use, etc.).
