Title
A GWAS Study of Metastatic Disease
Summary
We propose an analysis of existing GWAS data from cases across numerous cancers to identify susceptibility alleles associated with the risk of metastatic disease. The analysis will not focus on prognostic factors, but instead focuses only on the presence/absence of metastases, given the dominant nature of this factor on survival. Identification of genetic determinants of metastatic disease could provide insight into mechanisms of carcinogenesis and be of great importance, since the identification of geentic factors associated with susceptibility or resistance to disease progression could provide important clues to new therapies. Current, metastatic disease of the vast majority of solid tumors (stage 4) remains refractory to treatment.
Aims

There is only ONE core aim. Determine if there is a genetic component to metastatic disease across diverse malignancies. Metastatic disease is determined from stage data available for most tumors studied in PLCO.
