Title
Genetic Variation and Insulin-like Growth Factor Levels
Summary
The insulin-like growth factor pathway plays a key role in regulating somatic growth, and evidence suggests that it may contribute to the development of cancer. Inter-individual differences in IGF-1 and IGFBP-3 levels are thought to be partially determined by genetic factors; however, studies investigating the association between common polymorphisms and IGF serum levels have been limited in scope. We plan to comprehensively examine the association between common genetic variants and IGF-1 and IGFBP-3 serum levels using previously measured IGF levels and the genetic data available from the CGEMS genome-wide scan (GWAS) on the PLCO prostate cancer cases and controls. Results from the PLCO GWAS will then be pooled with those from the NHS in a meta-analysis, and any interesting findings will be replicated within other Harvard cohorts and/or DCEG cohorts.
Aims

The specific aims of this study are to: 1) examine the association between genetic variants genotyped in the CGEMS PLCO genome-wide scan and serum levels of IGF-1 and IGFBP-3; 2) conduct a meta-analysis of IGF-1 and IGFBP-3 association results from the PLCO and NHS genome-wide scans.
