Title
Interactions, mediating effects, and nonlinear effects of biological markers, genomics and environmental exposures on the occurrence and prognosis of various types of cancers
Summary
Although numerous studies have explored the influencing factors of the occurrence and prognosis of cancer, the interactions, mediating effects, and nonlinear effects of these factors has not been thoroughly explored. Therefore, this proposal aims to explore the impacts, especially interactions, mediating effects, and nonlinear effects, of biological markers, genomics and external environmental exposures (lifestyle, diet, etc.) on the occurrence and prognosis of various types of cancer, based on the cohort established from the PLCO trial. To achieve this goal, we look forward to collecting information of questionnaire, screening, laboratory testing, GWAS, etc. The additive and multiplicative interactions between influencing factors will be explored. Generalized additive models and restricted cubic spline functions will be used to examine nonlinear relationships. We will also explore how genotype and environmental exposures influence the occurrence and prognosis of cancer through biomarkers, by path analysis.
