Title
Evaluation of effects of dietary factors on the risk of colorectal cancer
Summary
Emerging evidences have demonstrated that both external exposure factors (e.g., dietary behaviors and colonoscopic surveillance) and internal exposure factors (e.g., vitamin D and hormone levels) could play important roles in the development and progression of colorectal cancer. In our previous study, we have found that individuals with a high level of BMI and estrogen had a higher incidence of colorectal cancer, whereas intake of aspirin, vitamin C, vitamin E and calcium supplements could reduce colorectal cancer risk (All these findings have not yet been published). Additionally, it has not reported that the PSA level be as a potential mediated factor on colorectal cancer risk. Besides, it seldom reported the interaction and mediation effect among multiple factors on colorectal cancer risk. We plan to use interaction analysis or/and mediation analysis to explain the effect of each risk factor on colorectal cancer, mainly including the potential factors of dietary behaviors, colonoscopic surveillance, multi-vitamins intakes and PSA level, and even the genetic variants. All these will helps to provide more comprehensive prevention strategies for colorectal cancer.
Aims
1. Investigate the association between dietary behaviors, colonoscopic surveillance, vitamin D intake, PSA level and genetic variants and colorectal cancer risk in the PLCO and in-house data.
2. Explore the interaction or mediation effect of each factor on colorectal cancer risk by using interaction and mediation analysis.
3. Provide the evidences to utilize more laboratory-based studies to explore the biological mechanisms of colorectal cancer.
